Summary :
Leptin and resistin are two members of a growing family of adipocytokines. Leptin is thought to regulate body fat, feeding behaviour, energy balance, fertility and neuroendocrine responses; whereas resistin antagonizes insulin action, impairs glucose tolerance and its activity is modulated by PPAR gamma receptors. It was previously shown that leptin is expressed in the rat hypothalamus and pituitary. With regards to resistin, we knew that PPAR-gamma receptors are expressed in the brain and pituitary. We also knew that ablation of the hypothalamus in mice led to obesity, insulin resistance and hyperglycemia. Here we go a step further in answering the questions of whether leptin is developmentally regulated, whether resistin protein is expressed in the mouse brain and pituitary, and what the role of the hypothalamus is in resistin expression.
Learning objectives :
The participant will acquire new knowledge about the regulation and expression patterns of leptin and resistin:
- Leptin mRNA, protein and receptors are developmentally regulated in the pituitary.
- Leptin receptors and protein are both expressed in the hypothalamus and pituitary, and are independently regulated.
- Resistin is expressed in the mouse brain and pituitary, along with PPAR – gamma receptors.
- Pituitary resistin is also developmentally regulated.
- Pituitary resistin expression is dependent upon an intact hypothalamus.
