Summary :
In this presentation Dr. Jandeleit-Dahm describes the Peroxisome Proliferator-Activated Receptor (PPAR) isoforms and their distribution in different tissue types, and presents data from experimental studies focusing on the investigation of vascular and anti-atherosclerotic effects of thiazolidinediones, which are PPAR gamma agonists.
Previous animal studies have demonstrated anti-atherosclerotic effects of thiazolidinediones with an improvement in insulin resistance. The apolipoprotein E knockout (ApoE KO) mice - control and diabetic - have been used by Dr. Jandeleit-Dahm's group in order to study the vascular and anti-atherosclerotic effects of PPAR gamma ligands independently of their insulin-sensitising effects. Preliminary data suggest that rosiglitazone may have direct vascular and anti-atherosclerotic effects, and some effects of PPAR gamma ligand treatment were also observed in the kidneys of these mice.
ApoE KO mouse studies have also indicated an important role of the vascular renin-angiotensin system in the development and acceleration of diabetes-associated atherosclerosis.
