Summary :
In this presentation Dr. Prins discusses the effects of TZDs on bone formation/resorption. Interestingly, these effects are partly caused by a TZD-induced increase in adipogenesis (1); the endocrinology of this phenomenon is discussed as well.

TZDs have an effect on many hormones including adiponectin, osteocalcin and TNF alpha. They were also shown to modulate the ratio of OPG and RANKL (2) leading to an increased activation of osteoclasts and bone resorption.

From the genetic point of view, Dr. Prins completes his presentation with some of the most recent results obtained with knockout mice (3). Some effects of TZDs on bone are beneficial and some detrimental, and it is not clear which is the major mechanism at work.
